摘要
目的:克隆并分析从阴道毛滴虫中得到两种蛋氨酸裂解酶同工酶基因(TVmgl1,TVmgl2)。方法:提取阴道毛滴虫总RNA,由逆转录获取cDNA,PCR大量扩增得到TVmgl1和TVmgl2基因,分离纯化后,连接pMD-19-T克隆载体,转菌后筛选得到阳性克隆,测序得到碱基序列。结果:序列分析所得基因,序列TVmgl1为1191bp,TVmglL2为1197bp,其编码氨基酸序列TVMGL1与TVMGL2有69%的同源性。结论:克隆得到TVmgl1、TVmgl2基因。
Objective: To colone, analyse the structure of the two genes encoding methionionase from Trichomonas vaginalis . Me.theds: Total RNA was extracted from primitive protozoon Trichomonas vaginalis,cDNA fragments encoding Methionine γ- lyase genes were amplified by RT-PCR using specific primers,and then cloned into pMD-19-T vector.The two inserted Methioninase genes were sequenced . Results: Analysis indicates that the DNA fragments are 1191bp and 1197bp in length and have 69% high nucleotide homology with that described in GenBank. Conclusion: Two L- Methioninc γ-lyasc isozyme genes were successfully cloned and identified from Triehomonas vaginalis.
