摘要
目的分析广西、云南和河南三地旋毛虫核糖体大亚基(mt-lsrRNA)基因差异,确定三个分离株的系统发生关系。方法分别提取三地旋毛虫分离株基因组,特异性扩增mt-lsrRNA基因。对三地旋毛虫及Genbank已知的旋毛虫相应基因序列进行对比研究。构建系统发生树,分析4株旋毛虫系统发生树的拓扑结构,同时收集三地的地理气候等信息,分析三地旋毛虫系统发生关系。结果广西旋毛虫分离株和云南旋毛虫分离株、河南分离株的mt-lsrRNA基因高度相似,在407个碱基对中,三地旋毛虫分离株与Genbank中T.spiralis序列相比,基因序列相似度很高,三个分离株在同一个位点发生T-C变异,此外,河南旋毛虫分离株在376位发生1个T-A变异。构建的系统发生树显示,广西分离株与云南、河南分离株及Genbank中的T.spiralis在同一分枝,进化距离为0.0000,自引导值为100,高度同源。三地旋毛虫与其它种类旋毛虫分于不同分枝,相距较远。结论三地旋毛虫分离株均为同一虫种,即T.spirilas,虽然宿主、地理环境及气候等因素不同,但进化差异小,亲缘关系近。
Objective To analyze the genetic variation of large subunit ribosomal-RNA among Triehinella isolates from the three provinces of Guangxi,Yunnan, Henan and explore phylogenetic relationships. Methods Genome DNA of the three Trichinella isolates were extracted respectively and mt-lsr RNA gene were amplified as the DNA marker by PCR. The homologieal nucleotides of T.spirdis published were downloaded. All sequences were compared with each other and distance matrixes were estimated. According to the data, phylogenic trees were constructed and analyzed by maximal parsimony(M-P) method. Results High degree of homology was found among the four isolates. In 407 base pairs,transversional from T to C in the same position was found. Besides,while Henan isolate and T.spiralis with a transitional change from T to A at 376 site. High homogeneity were found among the three isolates and T.spiralis .All of them belonged to one branch in phylogenic trees with evolution distance of 0.0000 among each of them and a bootstrap value of 100,Other Trichinella species were classified into other clades, which indicated the three Trichinella isolates is the same species as Zspirolis. Conclusion Although the host, environment and climate of loci where isolates from are different,all the three Trichinella isolates are tT..spirilas. Little molecular diversity and close phylogenetic relationship has been observed in mt-lsr RNA gene.
