摘要
目的探讨注射用紫杉醇脂质体对人子宫内膜癌细胞系的体外杀伤作用。方法以不同浓度含紫杉醇脂质体的培养基作用于人子宫内膜癌细胞系Ishikawa细胞,并设对照(普通培养基),通过MTT、倒置显微镜观察细胞增殖状况及形态,流式细胞分析技术检测该药对细胞生长的影响。结果经不同浓度紫杉醇脂质体作用24~96h,细胞存活率显著下降(P〈0.05);镜下观察显示部分细胞皱缩变形,崩解坏死,细胞脱壁悬浮;流式细胞术结果显示细胞凋亡率逐渐增加,细胞周期各时相中,S期比例减少。结论紫杉醇脂质体可抑制子宫内膜癌Ishikawa细胞的生长。
Objective To investigate the killing effect of liposome encapsulated paclitaxel on endometrial carcinoma Ishikawa cells in vitro. Methods The human endometrial carcinoma Ishikawa cell line was cultured with different concentrations of the liquid form of liposome encapsulated paclitaxel. MTT assay, inverted microscopic observation and flow cytometry assay (FCM) were then used to monitor the growth and proliferation of the cell line. Results When treated with different concentrations of liposome encapsulated paclitaxel for 24 to 96 hours, Ishikawa cells survival rate decreased significantly (P〈0.05); degeneration and necrosis of Ishikawa cells were found under microscope. The rate of cell apoptosis was increased and the rate of S phase was reduced. Conclusion Liposome encapsulated paclitaxel can inhibit endometrial carcinoma cells.
