甘草次酸对K562细胞基质金属蛋白酶MMP-2、MMP-9表达的影响 被引量:3

摘要 目的:研究MMP-2、MMP-9在K562细胞中的表达及甘草次酸(GA)对其表达影响。方法:采用MTT法检测甘草次酸对K562细胞增殖抑制的作用;半定量RT-PCR法和免疫组化分别检测GA对K562细胞基质金属蛋白酶MMP-2和MMP-9mRNA及蛋白表达的影响。结果:K562细胞抑制率与甘草次酸浓度及用药后培养时间呈正相关。GA能抑制K562细胞MMP-2、MMP-9mRNA及蛋白的表达,并呈剂量相关性。结论:MMP-2、MMP-9在白血病的发生发展中可能起一定作用,GA可抑制其增殖可能与下调MMP-2、MMP-9mRNA及蛋白表达有关。 OBJECTIVE To investigate the expression of MMP-2 and MMP-9 in K562 and the likely effects of glycyrrhitic acid (GA) on the expression of MMP-2 and MMP-9.METHODS MTT assay was performed to examine the effects of glycyrrhetic acid on the proliferation of K562 cells. The expression of rnRNA and protein of MMP-2 and MMP-9 in the K562 cells treated by GA were assayed bysemi-quantitied RT-PCR and Immunohistochemical studies. RESISTS There was statistically significant defference in the inhibition rate between the control group and GA groups. GA could downregulate the expression of mRNA and protein of MMP-2 and MMP-9 in treated K562 cells. CONCLUSION MMP-2 and MMP-9 may play some role in pathological process of leukemia. GA can inhibit the expression of MMP2 and MMP-9, it may have some influence on leukemia therapy.
