针灸对慢性应激抑郁模型大鼠环核苷酸的影响 被引量:4

Effect of Acupuncture and Moxibustion on cAMP Level in Chronic Mild Unpredictable Stressors Depression Rats

摘要 目的:探讨针灸对慢性应激抑郁模型大鼠血清及不同部位脑区环磷酸腺苷含量的影响及治疗抑郁症的机理。方法:将70只雄性wistar大鼠随机分为正常对照组、抑郁模型组、生理盐水组、手针"百会、太冲"组、手针"膻中、内关"组、电针"百会、太冲"组、电针"膻中、内关"组、艾灸"百会、太冲"组、艾灸"膻中、内关"组、阿米替林组,每组7只。采用放射免疫分析法观察了慢性应激抑郁模型大鼠血清及不同部位脑区环磷酸腺苷含量。结果:在血清中,模型组与生理盐水组大鼠cAMP含量较正常对照组显著下降;手针、电针"百会、太冲"穴组血清中cAMP含量较模型组显著升高,差异显著,与正常组比较无显著差异,手针"百会、太冲"穴组与电针"百会、太冲"穴组比较亦无显著性差异;手针、电针"膻中、内关"穴组和艾灸两组与模型组相比无显著差异;血清中cAMP含量:手针组>电针组,"百会、太冲"穴组>"膻中、内关"穴组。在大脑皮层中,模型组与生理盐水组大鼠cAMP含量较正常对照组显著下降;手针、电针"百会、太冲"穴和"膻中、内关"穴均能显著升高皮层cAMP含量,与模型组比较有显著差异,与正常组比较无明显差异,4组间比较无显著差异;艾灸"百会、太冲"和"膻中、内关"穴组虽能升高皮层中cAMP含量,但与模型组比较无显著性差异;皮层中cAMP含量:手针组>电针组,"百会、太冲"穴组>"膻中、内关"穴组。在海马中,模型组与生理盐水组大鼠cAMP含量较正常对照组显著下降;手针、电针"百会、太冲"穴和"膻中、内关"穴均能显著升高海马中cAMP含量,与正常组比较无显著差异,4组间比较无显著差异;艾灸"百会、太冲"穴与"膻中、内关"穴组与模型组比较无显著性差异;海马中cAMP含量:手针组>电针组,"百会、太冲"穴组>"膻中、内关"穴组。在下丘脑中,模型组与生理盐水组大鼠cAMP含量较正常对照组显著下降;艾灸"百会、太冲"、"膻中、内关"穴组与手针、电针"百会、太冲"穴组均能显著改善下丘脑cAMP的低水平状态,与模型组比较有显著差异,与正常组比较无显著差异;手针、电针"膻中、内关"穴虽能显著提高下丘脑cAMP含量,但与模型组相比无显著差异;下丘脑中cAMP含量:艾灸组>手针组>电针组,"百会、太冲"穴组>"膻中、内关"穴组;阿米替林组在血清、皮层、海马和下丘脑中cAMP含量与生理盐水组均无显著差异。结论:针灸可使大鼠脑内及血清中的cAMP含量升高,进而改善抑郁症状。对第二信使系统的调节作用是针灸治疗抑郁症的机制之一。 Objective: In order to investigate the effects of acupuncture and moxibustion on cAMP level in the animal model of chronic stressed depression and the treating mechanism of depression with acupuncture and moxibustion.Methods:70 male wistar rats were randomly allocated to ten groups as follows: control,depression model,saline,acupuncture,electric acupuncture and moxibustion of points " baihui,taichong" and "tanzhong,neiguan" and amitriptyline.The changes of cAMP level were observed by redioimmunoassay.Results:In serum,compared with control group,the cAMP level in model group and saline group decreased significantly;The level of cAMP in acupuncture and electric acupuncture on "baihui,taichong" point groups are higher than that in model group,while the two groups haven't distinct differences;The level of cAMP in acupuncture and electric acupuncture on "tanzhong,neiguan" point groups and the two moxibustion groups haven't distinct differences,compared with model group;The level of cAMP in acupuncture group is higher than that in electric acupuncture group,the level of cAMP in "baihui,taichong" group increased significantly,compared with "tanzhong,neiguan" group.In cortex,compared with control group,the cAMP level in model group and saline group decreased significantly;The level of cAMP in acupuncture and electric acupuncture on "baihui,taichong" and "tanzhong,neiguan" point groups are higher than that in model group,while the four groups haven't distinct differences;The level of cAMP in moxibution on "baihui,taichong" and "tanzhong,neiguan" point groups are higher than that in model group,but they haven't distinct differences;The level of cAMP in acupuncture group is higher than that in electric acupuncture group,the level of cAMP in "baihui,taichong" group increased significantly,compared with "tanzhong,neiguan" group.In hippocampus,compared with control group,the cAMP level in model group and saline group decreased significantly;The level of cAMP in acupuncture and electric acupuncture on "baihui,taichong" and "tanzhong,neiguan" point groups are higher than that in model group,while the four groups haven't distinct differences;The two moxibustion groups haven't distinct differences,compared with model group;The level of cAMP in acupuncture group is higher than that in electric acupuncture group,the level of cAMP in "baihui,taichong" group is higher than the that in "tanzhong,neiguan" group.In hypothalamus,compared with control group,the cAMP level in model group and saline group decreased significantly;The level of cAMP in acupuncture and electric acupuncture on "baihui,taichong" point groups and the two moxibustion groups are higher than that in model group;The level of cAMP in acupuncture and electric acupuncture on "tanzhong,neiguan" point groups are higher than that in model group,but the groups haven't distinct differences.The level of cAMP in moxibustion group is higher than that in acupunture group,while acupuncture group is higher than electric acupuncture group.The level of cAMP in "baihui,taichong" group increased significantly,compared with "tanzhong,neiguan" group.The level of cAMP in amitriptyline group has no distinct difference with saline group in serum,cortex,hippocampus and hypothalamus.Conclusion:Acupuncture and moxibustion can increase the serum and brain's cAMP of depressed rats.Regulating the second messenger system is the mechanism of acupuncture and moxibustion treatment.
