摘要
目的研究负载树突状细胞诱导实体肿瘤凋亡作用,为其用于临床生物治疗提供依据。方法从小鼠骨髓诱导培养树突状细胞,以CT-26肿瘤细胞冻融液为DC负载抗原,测定此负载的DC体内诱导CT-26实体肿瘤凋亡效果。结果诱导的树突状细胞形态典型;CT-26细胞冻融液负载的DC诱导实体瘤外周细胞凋亡明显,而内部细胞罕见凋亡。结论负载DC治疗体积较大的实体瘤作用有限,可能更适合微瘤治疗。
Objective ： To investigate the apoptosis effects induced with tumor antigen loaded dendritic ceils （DCs） and provide evidences for biological therapy based on DC. Methods： The freeze - lysate solution of CT - 26 tumor cells was loaded on DC that cultured from BALB/c murine bone marrow, and we detected the effect of apoptosis on solid tumor induced with DCs in vivo. Results： The outside cells of solid tumor could be induced apoptosis with antigen loaded DCs, while inside ceils of solid tumor had little apoptosis. Conclusion ： The effects of therapy on micro solid tumor with antigen loaded DC are potentially effective but not suitable for large solid tumor.
