缺氧复合烧伤血清刺激对新生大鼠心肌细胞粘弹特性影响的初步研究 被引量:2

摘要 目的:了解烧伤后因缺氧、烧伤血清对心肌细胞骨架的损伤机制及由此而产生的损伤效应和意义。方法:通过心肌细胞骨架免疫组化、骨架蛋白流式细胞术及心肌细胞粘弹性测定技术,动态观察体外培养乳鼠心肌细胞致伤前后的变化。结果:大鼠严重烧伤早期,即可引起心肌细胞弹性下降,骨架蛋白的荧光强度降低,中间丝及部分微管阳性信号减弱,培养心肌细胞呈脱落前征象。结论:心肌细胞骨架对维持心肌细胞的完整性是必需的,细胞骨架损伤可引起细胞脆性增加,细胞粘弹性下降,导致细胞生物力学特性改变,而这种变化可能直接参与烧伤后心肌损害的发生发展和转归过程。 Objective: To explore the mechanism of the injury of the cytomembrane and skeleton of cardiac muscle cells caused by the two injurious factors of hypoxia and burn serum after burns. Methods: The changes of the myocardial cells in the newborn rats were dynamically observed with immunohistochemistry, cytometry and measurement of cytomembranous viscoelasticity. Results: In the early stage after severe burns, the elasticity, the fluorescent intensity of the skeleton protein and the number of the positive signals of the intermediate filament and some microtubules were markedly decreased. Conclusion: The skeleton of the myocardial cells is essential to the integrity of its cytomembrane. The changes of the biomechanic characteristics of the cytomembrane resulted from the increase of cellular fragility and decrease of cytomembranous viscoelasticity caused by injury of the cellular skeleton might directly participate in the occurrence, development and improvement of the myocardial injury after burns.
