摘要
目的观察蛋白激酶C-α(PKC-α)在兔自体静脉移植中的动态表达,探讨其与移植血管内膜增生的关系。方法采用兔自体颈静脉移植模型,25只新西兰大白兔随机分为5组,其中手术Ⅰ~Ⅳ组分别于术后3,7,14,28d取材,假手术组(SO组)作为对照组。应用Real-TimePCR法、Western Blot法分别检测各组中PKC-α mRNA及其蛋白的表达变化;应用免疫组织化学法检测各组平滑肌细胞中PKC-α的表达。结果与对照组比较,PKC-α mRNA及其蛋白在术后3d时,表达均明显升高(P<0.01),而后逐渐降至正常。免疫组织化学检测PKC-α结果与Western蛋白印迹法一致。结论PKC-α可能参与了自体静脉移植后的血管平滑肌增生过程中的信号转导。
Objective To observe the expression of PKC-α in rabbits' autogenous vein graft and its relationship with the intimal hyperplasia. Methods Twenty-five New Zealand rabbits were randomly divided into five groups, Ⅰ,Ⅱ,Ⅲ,Ⅳ and SO group. The right common jugular vein was transplanted to the contralateral carotid artery. The specimen was collected at 3, 7, 14,28 d after operation. SO group was sham operation. In each group, the PKC-α mRNA and protein were detected by Real-Time PCR and Western Blot. Immunohistochemistry was also used to detect the PKC-α protein. Results Compared with SO group, the PKC-α mRNA and protein expression increased significantly at 3 d after operation, then returned to a normal state gradually. Oonclusion PKC-α might be involved in the signal transduetion of the vascular smooth muscle proliferation in autogenous vein graft.
