干扰素对小鼠α2(Ⅰ)型胶原基因启动子活性的影响 被引量:9

摘要 目的讨论干扰素的抗胶原蛋白合成作用的机制.方法以含小鼠胶原α2(Ⅰ)基因上游04kb,08kb,20kb启动片段为研究靶序列,应用CAT作报告基因,结合基因转染技术讨论IFNα,IFNγ对胶原基因的启动抑制活性.结果IFNα,IFNγ单独应用对小鼠胶原α2(Ⅰ)基因上游不同长短启动序列转录活性无阻断作用,但IFNα,IFNγ与TNFα联合应用则具有较强的抑制活性.结论IFNα,IFNγ的抗纤维化活性与其对小鼠α2(Ⅰ)胶原-2kb~+54bp启动活性的作用无直接相关,但IFNα,IFNγ可协助TNFα的抑制活性. AIM To study the effect of interferon on the promoter activity in mouse α2(Ⅰ) procollagen gene. METHODS The upper stream 400bp, 800bp, 2000bp sequences in mouse α2(Ⅰ) procollagen gene 5'flanking region were studied with CAT as reporter gene and gene transfection method. RESULTS IFNα and IFNγ had no inhibitory effect in various length promoter sequences of mouse α2(Ⅰ) procollagen gene, while the combination of IFNα,IFNγ and TNFα exerted obvious inhibitory effect. CONCLUSION The anti fibrotic effect of IFN might have no direct relationship with its action on promoter sequence of mouse α2(Ⅰ) procollagen gene. But the interferons have inhibitory effect on the promoters only when they are used in combination with TNFα.
