摘要
目的 利用基因芯片技术探讨胰岛素抵抗高血压大鼠基因表达谱及糖脂消治疗后的变化 ,研究胰岛素抵抗的分子发病机制及糖脂消治疗胰岛素抵抗高血压的作用机制。方法 用高果糖饲料诱发SD大鼠胰岛素抵抗模型 ,给予糖脂消口服后 ,用基因芯片分别检测高果糖组 ,治疗组 ,计算机软件分析后 ,观察基因表达的变化。结果 模型组表达差异的基因有 14 0条 ,新基因有 2 2条 ,已知基因 6 1条。治疗组表达差异的基因有 95条 ,新基因有 2 3条 ,已知基因 34条。结论 利用基因芯片技术检测胰岛素抵抗表达谱 ,证实糖脂消治疗可以改变其基因表达谱 。
Objective to discuss gene expression profile of insulin-resistance hypertensive rat and being treated by Tang-Zhi-Xiao using cDNA microarrary which could lead support to elucidate the molecular mechanisms underlying insulin resistance and Tang-Zhi-Xiao.Methods male Spraque-Darley rats were fed on high fructose diet to establish insulin resistant hypertensive rat model and taken orally Chinese traditional medicine, examined gene expression of high fructose control (FFR) and treatment control(FFR+TZX) using gene chip. Results a total of 140 genes, including 22 novel genes in FFR and that of 95 genes, 23 novel genes, were found.Conclusions Tang-Zhi-Xiao may induce gene changes and using this ways may offer a new means to discover mechanism of insulin resistance and Chinese traditional medicine.
