猪催乳素受体基因3′-UTR多态性及其与产仔性能的关联分析

Polymorphisms within 3′-UTR of PRLR Gene and its Association with Litter Size in Swine

摘要 通过研究PRLR基因多态性与母猪产仔性能的关系,为该基因在标记辅助选择中的应用提供依据。采用PCR-SSCP技术检测了大白猪、长白猪、马身猪、山西黑猪和山西白猪5个品种(系)共计323个个体PRLR基因3′-UTR的多态性.并采用单变量动物模型,分析了多态位点对母猪产仔性能的影响。结果表明,PRLR基因3′- UTR存在多态性,在所研究的群体中检测到A、B两个等位基因和AA、AB、BB三种基因型。在马身猪和山西白猪中,A等位基因占优势,频率分别为0.97和0.88;而在大白猪、长白猪和山西黑猪中,A、B两个等位基因的频率基本相似,分布在0.47~0.53之间。PRLR基因3′-UTR多态位点对母猪产仔性能无显著影响(P>0.05),不同基因型母猪的头胎产仔数和产活仔数均没有显著差异,但均表现出AB>AA>BB的趋势。 The aim of this study was to investigate the association of the polymorphism detected at porcine PRLR gene locus with the litter size in swine, and provide the basic knowledge for the PRLR gone as a genetic marker used in marker assisted selection. A total of 323 pigs from Large White, Landrace, Mashen, Shanxi Black and Shanxi White were used to detect the polymorphism within the 3′ untranslated region (UTR) of PRLR gene by PCR-SSCP, and the univariate animal model was used to analyze the effects of the PRLR genotype on the total number born (TNB) and number born alive (NBA) at first parity. The results showed that there was a polymorphic site within the detected region. Two alleles of A and B, and three genotypes of AA, AB and BB, were found in the examined populations. The A allele was the superior allele in Mashen (0.97) and Shanxi White (0.88), whereas the frequencies of A allele in Large White, Landrace and Shanxi Black were similar to those of B allele, with the frequencies ranging from 0.47 to 0.53. There was no significant association between the polymorphic site and the litter size of sows. The differences of TNB and NBA at first parity of sows with different genotypes, with the trend of AB〉AA〉BB, were not significant (P〉0.05).
