摘要
通过Linker连接呼吸道合胞病毒(RSV)G蛋白的部分抗原决定簇基因与大肠杆菌热不稳定肠毒素B亚单位基因(LTB)融合,构成融合基因,构建原核表达重组质粒并进行表达,并初步研究其免疫反应.构建得到的重组表达质粒,转化大肠杆菌后融合蛋白在大肠杆菌中得到了表达,用SDS-PAGE和Western-blotting对表达产物进行鉴定,融合蛋白的分子量约为25 ku.该蛋白能够被LT特异性抗体识别.成功表达呼吸道合胞病毒部分G蛋白与LTB的融合蛋白,并且该蛋白有一定的免疫原性.
To investigate the immunogenicity of recombinant mucous membrance vaccine of respiratory syncytial virus, a fusion protein gene was constructed by linking G: 130 ~ 230 and LTB gene with a linker and expressed in E. coli. The recombinant plasmid was transferred into E. coli and target protein was expressed in E. coli. The fusion protein was determined by SDS - PAGE and western - blotting. The molecular mass of fusion protein was estimate to be 25 ku. The fusion protein can be specificly recognized by LT antibody.The fusion protein showed certain immunity.
