摘要
本文报道一种从新鲜、冰冻猪脑垂体提取高纯度生长激素 (FGH) 的程序,包括常规的萃取、离子交换树脂层析和凝胶过滤。使用超速离心、聚丙烯酰胺凝胶电泳和生物学分析测定PGH的特性,其蛋白质的分予量是22000,沉降系数 (S20W) 为2.2267。每克新鲜猪脑垂体可获得0.8mg PGH。
The present work describes a procedure for the isolation of a highly purifed growth hormone from fresh-frozen porcine pituitary glands.The process Consists of conventional extraction,ion-exchange chromatography and gel filtration.The porcine growth hormone was examined for eharacterization by ultracentrifugation,polyacrylamide gel electrophore- sis and biological assays.It is a protein with a molecular weight of 22000 and the sedi- mentation coefficient(S20w)is 2.2267.Procine growth hormone was obtained in a yield of 0.8mg per gram fresh weight of porcine pituitary gland.
