摘要 研究了A.xylinumHN001菌株的特性.结果表明该菌株为革兰氏阴性,较好的碳源为甘露醇、山梨醇,蔗糖次之.菌株的最适生长和分泌纤维素温度为27~33℃;最适生长繁殖pH为5.0;分泌纤维素最适pH为3.9~4.5.最佳种子培养基的配比为:椰子水900g/L,(NH4)2SO42g/L,Mg-SO40.5g/L,蔗糖30g/L,pH为5.0;最佳生产培养基的配比:椰子水400~500g/L,(NH4)2SO45g/L,KH2PO40.5g/L,MgSO40.5g/L,NaAc2g/L,蔗糖40g/L,pH为3.9~4.2.其中椰子水用量为关键因子.菌株长期保藏宜采用冻干管. In this paper the characteristics of A.xylinumHN001 strain was studied. The result indicated that the strain is gram negative fungus. Its carbon comes from mannitol, soubitol, sucrose takes second place. The temperature of the best growth and secretion of cellulose is 27-33℃, the most suitable pH for growth and reproduction is 5.0, most suitable pH for secretion of cellulose is 3.9-4.5. The best allocated proportion (g/L) of seed culture medium is: coconut water 900, (NH4)2SO42, MgSO4 0.5, sucrose 30, pH value 5.0. Best allocated proportion (g/L) of production culture medium is : coconut water 400-500, (NH4)2SO4 5, KH2PO4 0.5, MgSO4 0.5, NaAc 2, sucrose 40, pH value 3.9-4.2. Coconut water amount used is most essential factor. The strain preserves for a long time suitably uses the freezing and drying tube.
