In order to investigate the expression and regulation of human heart chymase gene in mammalian cardiac tissue,the neonatal rat myocardial and nonmyocardial cells in cardiac tissue were cultured and transfected with plasmid pCH1,pCH2(containing 6 kb and 11 kb human heart chymase genes,respectively) and pBS KS respectively according to the modification of the calcium phosphate precipitation method.After total RNA was isolated,RNA Dot blotting was carried out with 0.8 kb human heart chymase cDNA as the probe.The results showed that both human heart chymase genes(6 kb and 11 kb)expressed only in cultured nonmyocardial cells.The results indicated that the expression of human heart chymase gene was highly cellular specific and provided for further researches in the function, in vivo ,of chymase gene.
