摘要
[目的]预测Pla a 1抗原的免疫原性及B细胞表位。[方法]联合运用多种方法预测Pla a 1的二级结构并预测其表面特性,如亲水性、可塑性及抗原表位。[结果]该蛋白是一个分子量约19 kDa,pI值约8.7,主要为α-螺旋的结构紧凑的球形蛋白。预测其亲水性区域:32-42,55-59,76-78,88-89,92-98,110-112,118-127,140-151,157-163;预测其可塑性区域:28-40,49-57,76-80,87-98,109-114,121-125,137-139,144-151,154-162;预测其蛋白质抗原表位区域:29-42,49-60,75-78,86-100,107-114,116-130,134-152,156-164;预测其转角结构区域:39-42,51,55,88-89。[结论]Pla a 1抗原蛋白具有较强的免疫原性;其主要的抗原表位集中于29-42,49-60,86-100,而这些预测结果为进一步寻求梧桐花粉变态反应的防治方法提供了依据。
[ Objective ] To predict the immunogenicity and B cell epitopes for Pla a 1 antigen. [ Method ] Secondary structure and tertiary structure and surface properties of Pla a I , such as hydrophilicity and plasticity were predicted with various methods. [ Results ] Pla a 1 was predicted to be a global protein with molecular weight about 19kDa and with pI value about 8.7. And it showed a predicted secondary structure characteristic of all-alpha proteins. By computation, Many hydrophilicity regions in pla a 1 were identified: 32-42, 55-59, 76-78, 88-89, 92-98, 110-112, 118-127, 140-151, 157-163; many plasticity regions in pla a 1 were identified: 29-42, 49-60, 75-78, 86-100, 107-114, 116- 130, 134-152, 156-164; many antigenic epitopes in pla a 1 were identified:29-42, 49-60, 75-78, 86-100, 107-114, 116-130, 134-152, 156-164; many turn regions in pla a 1 were identified: 39-42, 51, 55, 88-89. [ Conelution] Pla a 1 has a strong immunogenicity. And its B cell epitopes are mostly localted on 29-42, 49--60, 86-100, therefore, which can be useful for diagnosis and therapy.
