脐带间充质干细胞分离培养和生物学特性的初步研究 被引量:2

摘要 目的建立从脐带分离间充质干细胞的分离培养方法,探讨研究其基本生物学特征.方法无菌条件下收集健康孕妇剖宫产的新生儿脐带,用酶消化法获取间充质干细胞(mesenchymalstemcells,MSCs),经传代培养、倒置显微镜观察形态、细胞增殖能力观察及流式细胞仪检测脐带间充质干细胞免疫表型.结果经传代培养后,脐带间充质干细胞呈长梭形、旋涡状生长.细胞免疫表型显示,CD34,CD45,CD14阴性;CD90,CD73,CD105阳性.结论脐带间充质干细胞与骨髓来源的间充质干细胞形态无明显变化,且免疫表型类似,是一种新的间充质干细胞来源. Objective To establish the method of separate culture to separate mesenchymal stem cells from umbilical cord for exploring and investigating its basic biological speciality. Methods Under sterile conditions, to collect the umbilical cord of newbron from healthy pregnant woman. To get mesenchymal stem cells (MSCs) with the method of enzyme digestion,and to experience subculture, morphologic observation with inverted microscope and observation of faculty for cell reproduction. And with flow cytometer to test the immunophenotype of mesenchymal stern cells in umbilical cord. Results After subculture,the mesenchymal stem cells of umbilical cord showed the status of long,fusiform and eircinate growth. The immunophenotypes of cells were that CD34,CD45 and CD14 was negative;CD90,CD44 and CD105 was positive. Conclusion There is no distinct change between mesenchymal stem cells of umbilical cord and mesenchymal stem cells of spinal cord. And they have similar immunophenotype. The mesenchymal stem cells of umbilical cord is a new origianl for mesenchymal stem cells.
