新型重组人肿瘤坏死因子抗荷鼻咽癌裸鼠的实验研究 被引量:5

摘要 目的寻找理想的鼻咽癌生物治疗及联合治疗的新方法。方法利用国内建株的鼻咽癌细胞株(CNE-2)制造了裸鼠鼻咽癌模型,用新型重组人肿瘤坏死因子(NrhTNF-α),通过不同的用药途径(局部、全身)和模式(单独、联合)观察治疗荷鼻咽癌裸鼠的疗效。并在光镜和透射电镜下观察NrhTNF-α作用后肿瘤组织细胞组织病理学及超微结构,探讨其作用机理。结果和结论①荷鼻咽癌裸鼠经NrhTNF-α治疗后可出现肿瘤出血、坏死,肿瘤缩小、消失、生存期延长。②NrhTNF-α局部使用的抗瘤效果优于全身用药。③NrhTNF-α与Cab联合使用具有协同作用,增强抗瘤效果。④组织病理学及超微结构观察发现NrhTNF-α首先引起细胞质内线粒体、内质网肿胀、整个细胞空泡样变性,逐步导致核浓缩,崩解,最终导致细胞死亡。该研究为临床上鼻咽癌生物治疗及联合治疗提供实验基础。 Objective Nude mice model bearing nasopharyngeal carcinoma were established using nasopharyngeal squamous carcinoma cell line ( CNE 2) . Methods New recombinant human tumor necrosis factor (NrhTNF α) and carboplatin were administered locally or systemically,single or in combination. The microstructural and ultra micro structural changes of carcinoma after administration of NrhTNF α were oberserved under light and electron microscopes. Results and Conclusion ①After treatment, the tumor showed hemorrhage、necrosis、regression, the survival time was prolonged. ②Locally administered NrhTNF α had more significant antitumor effect than intravenously administered of NrhTNF α. ③There is a notable synergistic antitumor effect by the combination use of NrhTNF α and carboplatin. ④Microscopy showed that cell changes mostly took place in plasma especislly in mitochondria and endoplosmic reticula. The mitochondria were highly bloated. Besides, some nuclei were condensed and fragmented, resulted in cell death. These studies provide experimental basis for biological treatment of nasophayngeal carcinoma.
