摘要
目的探讨脉络膜新生血管(choroidal neovascularization,CNV)生成过程中环氧化酶-2(cyclooxygenase-2,COX-2)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达及其意义。方法532nm激光诱导棕色挪威(Brown-Norway,BN)大鼠实验性CNV。光凝后7d,采用免疫组织化学法检测COX-2和VEGF在CNV中的表达。采用Western blotting和RT-PCR方法检测光凝后1d、3d、7d和14d CNV中COX-2和VEGF蛋白与mRNA表达的变化。结果CNV区域的血管内皮细胞和基质细胞均有COX-2和VEGF蛋白的表达,主要表达于胞浆中。COX-2蛋白及其mRNA的表达在光凝后3d时达高峰,VEGF蛋白及其mRNA的表达在光凝后7d时达高峰。COX-2蛋白及其mRNA的表达早于VEGF蛋白及其mRNA的表达。结论COX-2在CNV生成早期表达上调,且其表达先于VEGF的表达,提示COX-2可能通过调控VEGF的表达在CNV生成过程中发挥重要作用。
Objective To investigate the expression and their significance of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) in choroidal (CNV). Methods The experimental CNV was induced by 532 nm laser photocoagulation in Brown-Norway rat. The expression of COX-2 and VEGF in CNV were detected by immunohistochemistry at 7 days after photocoagulation. The protein and mRNA of COX-2 and VEGF were examined by Western blotting and RT-PCR at 1 day,3 days,7 days, 14 days after photocoagulation,respectively. Results The protein of COX-2 and VEGF were found in vascular endothelial cells and stromal cells in CNV, and were most prominent in kytoplasm. The protein and mRNA of COX-2 reached the maximum at 3 days after photocoagulation, and those of VEGF reached the maximum at 7 days after photocoagulation. The expression of protein and mRNA of COX-2 was prior to those of VEGF. Conclusion COX-2 increases in the early stage of experimental CNV and the expression of COX-2 is prior to the expression of VEGF, which indicates that COX-2 may play an important role in the pathogenesis of CNV by regulating VEGF.
