环氧化酶-2和血管内皮生长因子在激光诱导的大鼠脉络膜新生血管中表达的相关性 被引量:7

Correlation between expression of cyclooxygenase-2 and vascular endothelial growth factor in laser-induced choroidal neovascularization in rat

摘要 目的探讨脉络膜新生血管(choroidal neovascularization,CNV)生成过程中环氧化酶-2(cyclooxygenase-2,COX-2)和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达及其意义。方法532nm激光诱导棕色挪威(Brown-Norway,BN)大鼠实验性CNV。光凝后7d,采用免疫组织化学法检测COX-2和VEGF在CNV中的表达。采用Western blotting和RT-PCR方法检测光凝后1d、3d、7d和14d CNV中COX-2和VEGF蛋白与mRNA表达的变化。结果CNV区域的血管内皮细胞和基质细胞均有COX-2和VEGF蛋白的表达,主要表达于胞浆中。COX-2蛋白及其mRNA的表达在光凝后3d时达高峰,VEGF蛋白及其mRNA的表达在光凝后7d时达高峰。COX-2蛋白及其mRNA的表达早于VEGF蛋白及其mRNA的表达。结论COX-2在CNV生成早期表达上调,且其表达先于VEGF的表达,提示COX-2可能通过调控VEGF的表达在CNV生成过程中发挥重要作用。 Objective To investigate the expression and their significance of cyclooxygenase-2 (COX-2) and vascular endothelial growth factor (VEGF) in choroidal (CNV). Methods The experimental CNV was induced by 532 nm laser photocoagulation in Brown-Norway rat. The expression of COX-2 and VEGF in CNV were detected by immunohistochemistry at 7 days after photocoagulation. The protein and mRNA of COX-2 and VEGF were examined by Western blotting and RT-PCR at 1 day,3 days,7 days, 14 days after photocoagulation,respectively. Results The protein of COX-2 and VEGF were found in vascular endothelial cells and stromal cells in CNV, and were most prominent in kytoplasm. The protein and mRNA of COX-2 reached the maximum at 3 days after photocoagulation, and those of VEGF reached the maximum at 7 days after photocoagulation. The expression of protein and mRNA of COX-2 was prior to those of VEGF. Conclusion COX-2 increases in the early stage of experimental CNV and the expression of COX-2 is prior to the expression of VEGF, which indicates that COX-2 may play an important role in the pathogenesis of CNV by regulating VEGF.
