摘要
以草鱼和鲤为材料,提取基因组DNA,制备RAPD-PCR模板,对3个随机引物盒共60个10-mer引物进行了PCR扩增,共筛选出7个较理想的可用于鱼种内或鱼种间群体遗传分析的随机引物.用这7个引物所获得的草鱼和鲤的RAPD指纹图谱平均带纹总数分别为4.28±1.89和6.71±4.19,多态性带纹总数分别为1.71±1.60和5.14±4.74.RAPD指纹图谱特征反映出草鱼比鲤具有更高的遗传纯度.7个引物共扩增出草鱼特异性带纹18条,鲤特异性带纹32条.大量特异性带纹的检出表明,两个鱼种间存在较大的遗传差异。
Genomic DNA was extracted as RAPD PCR template from the liver tissue of grass carp and common carp, 60 10 mer random primers were used and 7 primers were proved to be ideal for population genetic analysis within and or between the fish species. The average number of total RAPD bands amplified with these 7 primers for grass carp and common carp were 4 28±1 89 and 6 71±4 19,respec tively, the average number of polymorphic bands were 1.71±1.60 and 5.14±4.74, respectively. The RAPD patterns suggested that the grass carp have higher genetic homogeneity than the common carp. 18 grass carp specific bands and 32 common carp specific bands were produced by all 7 primers, which implied significant genetic variation between the grasscarp and the common carp,and provided the candidate genetic markers for molecular assay of the gene transfer (especially whole genomic DNA transfer) between these two fish species.
