摘要
应用兔离体肺再灌注模型研究前列腺素E1(PGE1)及硝酸甘油(NTG)在离体肺保护中的作用。18只兔随机平均分为两组,对照组以LPD液进行肺灌洗及保存,用药组以PGE1100μgL-1及NTG10mgL-1加入LPD液中进行肺灌洗及保存。每组再分为三小组(各含6个肺)分别保存2、4、8小时后进行再灌注,测定再灌注后肺血管阻力(PVR),肺通气阻力(LR),肺含水量(LW)及动脉血氧分压(PaO2)以了解肺保护效果。结果示:用药组再灌注后PVR明显低于对照组(P<0.05)。LR、LW及PaO2两组无差别。说明PGE1及NTG在保护肺血管内皮细胞及降低再灌注后肺血管阻力方面明显优于对照组。PGE1及NTG作用机理可能是通过肺动脉扩张改善肺早期灌洗及抵制中性粒细胞。
Using isolatedlung reperfusion model, the effects of prostaglandin E 1(PGE 1) and nitroglycerin (NTG) on lung preservation were studied in 18 rabbits which were randomly divided into two groups (9 rabbits in each group). In the control group, the lungs were flushed and preserved with low potassium dextran (LPD) solution, while in the treated group, PGE 1(100 μg·L-1) and NTG (100 mg·L-1) were added to LPD solution to flush and preserve the lungs. Each group was then divided into three subgroups. Reperfusion was performed in each subgroup at 2 h, 4 h and 8h, respectively. Pulmonary vascular resistance (PVR), lung resistance (LR), lung water (LW)and blood gas after reperfusion were determined. It was found that: 1) PVR was much lower in the treated group than in the control group (P<0.05); 2) No difference was found in LR, LW and PO 2 between the two groups. It was concluded that PGE 1 and NTG can improved the lung preservation by protecting the pulmonary vascular endothelial cells and decreasing the PVR after reperfusion.
