人巨细胞病毒在体外新生乳鼠海马神经细胞中的增殖能力 被引量:2

Proliferative Ability of Human Cytomegalovirus in Hippocampal Nerve Cells of Neonatal Rats Infected in Vitro

摘要 目的探讨人巨细胞病毒(HCMV)AD169株在新生乳鼠海马神经细胞体外培养物感染情况及其是否存在增殖能力。方法用人胚成纤维细胞悬液培养增殖,按病毒致细胞病变效应终点稀释法以24~48 h出现(+++)~(++++)的病毒悬液在人胚成纤维细胞上滴定半数组织细胞感染量为5×107L-1,将病毒分别接种在SD大鼠长成单层的新生乳鼠海马48 h神经细胞培养物上,每天显微镜观察记录,采用免疫组织化学方法检测神经细胞培养物上巨细胞病毒早期蛋白,间接免疫荧光检测HCMV pp65蛋白,透射电镜方法观察和鉴定HCMV在神经细胞内的感染情况。结果接种病毒的海马神经细胞在感染后第1天即可观察到特征性细胞病变效应,随着时间的延长,细胞裂解液化消失;光镜下,免疫组织化学可见巨细胞病毒产生早期蛋白表达于受感染的培养海马神经细胞中,呈棕黄色颗粒;HCMVpp65蛋白免疫荧光检测亦为阳性,阴性对照未见任何阳性信号;透射电镜可见新生鼠大脑海马神经元胞质中有直径为80~120 nm病毒颗粒。结论HCMVAD169株能够感染乳鼠海马神经细胞,并具有一定的增殖能力。 Objective To explore infectious of human cytomegalovirus (HCMV) AD169 in neonatal marine hippocampal nerve cells in vitro and whether they have proliferative ability. Methods They were proliferatived with human embryonic fibroblasts(HF) cells,eytopathic effect was ( + + + ) - ( + + + + ) within 24 -48 hours,and 5 × 10^7 L^-1 cell suspension of the HCMV AD169 strain was inoculated onto monolayer nerve cell media of SD neonatal rat hippocampal nerve cells. Cytopathic effect was observed each every day with microscope ,early HCMV protein and pp65 protein were detected by immunohistochemistry and indirect immunofluorescent;the virus titers of cultured supernatant neonatal rats hippocampal nerve cells were determined, and HCMV particles in the nerve cells were observed by electron microscopy. Re- suits Characteristic cytopathic effect was observed on the first day of inoculation and this cytopathic change aggravated as inoculation time prolonged. HCMV early proteins were found using immunohistochemistry, they were buffy panicles in light microscope. HCMV characteristic protein pp65 was detected by indirect immunofluorescent examination and the controls were negative. HCMV particles were observable in the cytoplasm and karyon of the nerve cells under the election microscopy and the diameter of panicles was 80 - 120 nm. Conclusion HCMV can infect neonatal murine hippocampus nerve cells in vitro and has certain proliferative ability.
