摘要
目的探讨吗啡成瘾戒断后大鼠海马CA1区P-CREB的表达变化。方法48只健康雄性成年SD大鼠,随机分为实验组和对照组:实验组腹腔注射吗啡起始剂量5mg/kg,1d2次,逐日递增,连续10d;对照组:以生理盐水代替吗啡。停药后7d、14d和21d处死。用免疫组织化学方法(ABC法)检测海马CA1区P-CREB的表达,图像分析系统测定阳性反应产物的平均灰度值。结果P-CREB表达在7d、14d实验组与对照组相比表达上调(P<0.05),21d与对照组比较无显著差异(P>0.05)。结论海马CA1区CREB磷酸化在吗啡依赖的形成中发挥作用。
Objective To investigate the effects of morphine on the expressions of phosphorylated CREB (P-CREB) in CA1 area of hippocampus of the morphine dependent rats. Methods 48 healthy adult SD male rats were randomly divided into experimental and control groups : experimental rats were initially injected of morphine dose 5 mg/kg, twice a day, increasing daily 5 rag, for 10 days. The animals of control group were injected with NS instead of morphine. The animals were sacrificed on the 7, 14, and 21 d after stopping injection. P-CREB expressions in CA1 area of hippocampus were observed by immunohistochemical technique (ABC method), and the mean gray values of immunoreactive product was detected by image analysis system. Results the expressions of P-CREB had significant differences between experimental groups and control groups on 7 d, 14 d after stopping injection (P〈0.05). There were no differences of P-CREB expression on 21 d after stopping injection compared with control groups. Conclusion P-CREB expression in CA1 area of hippocampus may be one of the reasons for morphine dependence after morphine treatment in the adolescent.
