摘要
[目的]研究新疆雪莲分化过程中的生理生化变化。[方法]将以雪莲无菌苗的叶片为外植体诱导的雪莲胚性愈伤组织置于MS+2mg/LNAA+0.5mg/L6-BA(pH值为5.85~5.90)培养基上进行培养,对培养过程中雪莲细胞可溶性蛋白含量、可溶性糖、淀粉酶等参数进行测定分析。[结果]在新疆雪莲整个培养阶段,胚性愈伤纽织中的蛋白质含量高于非胚性愈伤组织,可溶性糖保持稳定,仅在芽分化阶段有一定的上升,α-淀粉酶活性保持稳定。胚性愈伤组织中的β-淀粉酶的活性均高于非胚性愈伤组织,这说明阻淀粉酶与细胞的分化过程密切相关。β-淀粉酶在整个培养过程中呈现双峰曲线。第1个峰出现在培养的开始阶段,第2个峰出现在芽分化时期。[结论]该研究对研究新疆雪莲的高频再生具有重要的指导意义。
[ Objective ] The aim was to study the change of physiology and biochemistry of Xinjiang Saussurea involucrata during differentiation. [ Method] The embryogenic callus of S. involucrata induced by the explant that was from the leaves of S. involucrata aseptic seedlings was put on the medium of MS + 2 mg/L NAA + 0.5 mg/L 6-BA(pH 5.85 - 5.90) to culture, the parameters of soluble protein content, soluble sugar and amylase etc. of S.involucrata cells during culture process were detemined and analyzed. [ Result ] In the whole culture process of Xinjiang S. involucrata, the protein content of embryonic callus were higher than those of non-embryonic ones, the soluble sugar content kept steady,only had some rise during bud differentiation;a-amylase ac- tivity kept steady. The β-amylase activity in embryonic callus was all higher than that of non-embryonic callus,which indicated that β-amylase had a close correlation with differentiation process of cells.β-amylase showed bimodal curve in the whole cnlture process,the first peak appeared at the beginning stage of culture,and the second peak presented at bud differerltiation phase. [ Conclusion] The study had important guiding significance in the research of high frequency regeneration of Xinjiang S. involucrata.
