摘要
目的观察不同活度的125I粒子植入对荷人纤维肉瘤裸鼠移植瘤的杀伤作用。方法建立荷人纤维肉瘤裸鼠皮下移植瘤模型32只,随机分为4组,移植瘤内分别植入为空源粒子,活度为14.8、22.2、29.6MBq粒子各1颗。术后15d,观察裸鼠存活率、皮肤放射性损伤、移植瘤体积及病理学变化。结果对照组、低、中、高活度组存活率分别为75.0%、83.3%、100.0%和100.0%;实验组、对照组皮肤反应,差异有统计学意义(P〈0.05);不同活度组抑瘤率差异有统计学意义(P〈0.05)。病理组织学反应程度:中、高活度组差异无统计学意义(P〉0.05),其他各组差异有统计学意义(P〈0.05)。结论125I粒子植入对荷人纤维肉瘤裸鼠移植瘤有显著抑瘤效果,粒子活度为影响抑瘤效果和皮肤损伤的因素。
Objective To investigate the effect of different activity of 125I radioactive seed implantation on nude mice with transplanted human fibrosarcoma. Methods Animal model simulating human fibrosarcoma was established by subcutaneous implantation of HT-1080 cell lines into nude mice. Mice were randomly divided into 4 groups:control group,low-activity group, moderate-activity group and high-activity group,which were respectively treated with different activity of 125I seed:0,14.8,22.2 and 29.6 MBq. Survival rate, regional skin reaction, tumor volume and pathological changes were detected respectively in all nude mice on the 15th day after the 125I seed implantation. Results The survival rate in control group,low-moderate and high-activity groups were 75.0% ,83.3% , 100% and100% respectively. There was significant difference in regional skin reaction between control and different activty groups (P 〈 0.05 ). There were significant differences ( P 〈 0.05 ) in the tumor growth inhibitory rate between low-activity group, moderate-activity group and high-activity group. There was significant difference in the extent of pathological response between control and different activity groups ( P 〈 0.05 ), except for the difference between moderate-activity group and high-activity group ( P 〉 0.05 ). Conclusion Interstitial implantation with 125I seed had significant inhibitory effects on the human fibrosarcoma xenografts in nude mice, and the activty of the 125I seed can effect treatment effectiveness and the extent of radiation injury.
