同种带瓣主动脉移植后影响供体组织钙含量的相关性研究 被引量:1

Correlative study of calcium content in rat aortic valved homograft tissue

摘要 目的探讨同种带瓣主动脉移植后影响供体组织钙含量的相关因素。方法将90只大鼠随机分为异基因组(冷冻组、治疗组)和同基因组。术后2、4、8、12、16周检测大鼠血液中T细胞抗原抗体(TCR)-α、β、CD28的表达。取出移植物,用光镜和电镜观察移植物组织结构形态学变化,用免疫组织化学染色方法检测动脉壁CD34的表达,用原子火焰吸收法测定同种带瓣主动脉(AVH)供体组织钙的含量。结果异基因组移植后TCR—α、β,CD28的表达水平均较同基因组明显增强(P〈0.01)。异基因组供体组织钙含量自移植后4周开始升高,同基因组各时点的钙含量差异无统计学意义(P〉0.05)。冷冻组于术后4、8周其供体组织钙含量(μg/g)为2856±79、3587±168;DC单抗治疗组为2176±210、3089±176;同基因组为860±60、870±50。结论移植物钙含量与免疫排斥反应密切相关,免疫抑制治疗可减轻免疫反应,延缓钙化进程。 Objective To investigate the corrrelative factors influencing calcium contert in rat aortic valved homograft tissue. Methods The study was assigned to allogenc and isogenenic groups. Recipient rats were randomly allocated into groups. Rat AVH with myocardial cuff were heterotopically allografted onto abdominal aorta of rats. The rats were sacrificed in batchs at 2,4,8,12 and 16 weeks postoperatively. Blood samples were obtained for detecting the expression of TCR-α and β, and CD28. The AVH specimens were obtained for observing the change of endotheliocytes and smooth muscle cells under the light and electron microscopy,and detecting the expression of CD34 by. At the same time, calcium contents of AVH tissues after transplantation were determined. Results The expression of TCR-α and β, and CD28 in allogene group was signficantly higher than in the isogenic group at each time point (P 〈 0.01 ). Calcium contents of AVH tissues in allogene group were increased gradually at 4th week, and no significant difference in calcium levels was found in isogenenic group over 5 different periods ( P 〉 0.05 ). In the cryopreservation group caleimn contents of AVH tissues were 2856 ± 79 and 3587± 168 respectively at 4th and 8th week postoperatively;2176±210 and 3089 ± 176 in DCmAb treatment group, and 860 ±60 and 870 m 50 in isogenic group. Conclusion Calcium contents of AVH tissues were closely correlated with immune rejection response. Imnmnosuppressive treatment could alleviate immune rejection response and delay the course of AVH calcification.
