摘要 目的观察SIRT1mRNA在成人急性白血病骨髓细胞中的表达及其与临床疗效之间的关系。方法应用半定量RT-PCR检测SIRT1mRNA在50例成人急性白血病骨髓单个核细胞中的表达情况。结果SIRT1mRNA的表达初治组、复发组均高于正常对照组(P<0.05),复发组高于初治组(P<0.05),AML组与ALL组的差别无统计学意义(P>0.05),SIRT1mRNA的表达与性别、年龄、初诊时白细胞计数及髓外浸润情况无关(P>0.05),SIRT1mRNA高表达组的完全缓解率低于低表达组(χ2=5.021,P<0.05)。结论SIRT1mRNA在成人初治及复发急性白血病骨髓细胞中高表达,可能参与白血病的发生发展及耐药。 [Objective] To investigate the SIRT1 expression in adult acute leukemia (AL) and its clinical significance. [Method] The SIRT1 expression were detected by aemi-quantitive reverse transcription polymers chain reaction (RT-PCR) in 50 adult AL patients and 15 normal controls. [Results] The expression of SIRT1 in 40 newly diagnosed AL patients and 10 AL patients in relapse were markedly higher than in 15 normal controls respectively (P 〈0.05). In relapsed patients,the expression of SIRT1 was higher than in newly diagnosed patients(P 〈0.05). There was no significant difference between newly diagnosed AML patients and ALL patients in SIRT1 expression (P 〉 0.05). SIRT1 expression was not related with gender, age,WBC and extramedullary infiltration (P 〉0.05). The CR rate of SIRT1 high expression group was lower than that of low expression group (P 〈0.05). [Conclusion] SIRT1 had higher expression in newly diagnosed and relapsed AL patients which may contribute to pathogenesis and drug resistance of AL
