附子对慢性充血性心力衰竭模型大鼠NO、TNF-α水平的影响 被引量:14

摘要 [目的]探讨附子抗心力衰竭的疗效及作用机制。[方法]用SD大鼠50只,随机分正常、空白对照组(简称Ⅰ号组)、模型组(简称Ⅱ号组)、地高辛组(简称Ⅲ号组)、附子水煎剂低剂量组(简称Ⅳ号组)、附子水煎剂高剂量组(简称Ⅴ号组)。除正常对照组外均采用尾静脉注射盐酸阿霉素(ADR)损伤大鼠心肌致心力衰竭。各组分别用药治疗后,测定大鼠血清中TNF-α、NO水平,并应用光镜观察心肌组织的理变化。[结果]与正常对照组比较,心衰模型组大鼠血清中TNF-α、NO水平升高;与心衰模型组比较,使用地高辛能显著降低心衰模型大鼠血清中TNF-α、NO水平,差异显著(P<0.01);而附子水煎剂高剂量、附子水煎剂低剂量则可轻度降低心衰模型大鼠血清中TNF-α、NO水平,两组无明显差异(P>0.05),但这两组跟地高辛组差异显著(P<0.01)。光镜结果显示附子水煎剂高剂量组、附子水煎剂低剂量组心肌细胞损伤度均轻于心衰模型组,而地高辛组心肌细胞损伤度明显轻于心衰模型组。[结论]附子高、低剂量组能轻微改善心功能、减轻心衰症状,调节神经内分泌细胞因子水平,必要时应复方使用,方能收到满意的疗效。 [Objective]To discuss the cure effect and mechanism of aconite anti-heart failure. [Method]Randomly divide 50 SD rats into normal, blank control group(Ⅰ), model group(Ⅱ), Digoxin group(Ⅲ), low-dosage(Ⅳ) and high-dosage group(Ⅴ) of aconite decoction. Except for normal control group, the others take tail vein injection of ADR to injure rats cardiac muscle for heart failure. All groups were tested serum TNF-α and NO levels after medication, and observe cardiac muscle change under microscope. [Result]Compared with normal control group, the TNF-α and NO were increased in model group; compared with model group, TNF-α and NO were decreased markedly after used with Digoxin, the difference was obvious (P〈 0.01 ); TNF-α and NO were slightly decreased in groups Ⅳ and Ⅴ, there's no marked difference between them(P〉0.05), but Ⅳ and Ⅴ had obvious difference with Ⅲ(P〈0. 01). The microscope showed Ⅳ and Ⅴ had less injured cardiac muscle than group Ⅱ, the injury in Ⅲ was markedly less than Ⅱ. [Conclusion]Groups Ⅳ and Ⅴ can slightly improve heart function, relieve heart failure symptoms, and regulate nerve endocrine cell factor level. If necessary, the compound of it can get satisfying result.
