摘要 目的探讨Kruppel样因子4(KLF4)在肝细胞癌(简称肝癌)中的表达及意义。方法应用寡核苷酸基因芯片、逆转录多聚酶链反应(RT-PCR)、免疫组织化学技术检测KLF4在健康人、肝硬化及肝癌组织中的表达,并分析KLF4与肝癌临床病理特征的关系。结果寡核苷酸基因芯片分析,KLF4在肝癌差异表达基因谱中明显下调(Ratio=0.438);RT-PCR及免疫组织化学方法证实,KLF4在肝癌中低表达,其表达缺失与病理学分级显著相关。结论KLF4在肝癌组织中的低表达与肿瘤分化程度密切相关。 Objective To explore the expression and significance of Kruppel-like factor 4 (KLF4) in the hepatocellular carcinoma(HCC). Methods Using the oligonucleotide chip, reverse transcriptional polymerase chain reaction (RT- PCR) and immunohistochemistry, the expressions of KLF4 was detected in tissues of normal human liver, cirrhosis and HCC. It's necessary to investigate the relationship between their expressions and their clinical pathological characteristics. Results KLF4 expression was significantly down regulated in HCC based on our microarray study( Ratio = 0.438), which was respectively confirmed by RT-PCR and immunohistochemistry analysis. The study shows that the deletion of KLF4 was highly correlated to pathology grading of HCC. Conclusions The low expression of KLF4 in HCC is intimately correlated to the extent of tumor differentiation.
