摘要
目的观察转化性生长因子β(TGF-β)对肝癌细胞的促进作用。方法 BUF大白鼠在接种7316A肝癌细胞的同时应用人γTGF-β_1;治疗,并于接种后5~20天测量肿瘤体积;γTGF-β_1还分别加入混合淋巴细胞培养(MLR)及7316A肝癌细胞培养中。结果大白鼠经TGF-β治疗组的肿瘤体积显著大于讨照组(P<0.01);TGF-β明显抑制T淋巴细胞的MLR反应(P<0.05),但对了316A肝癌细胞的增殖反应无明显抑制作用。结论由于TGF-β是肝再生过程产生的肝细胞再生抑制因子,提示肝硬化患者肝癌的发生率明显增高,可能与TGF-β抑制了免疫活性细胞的增殖,造成有利于肿瘤细胞生长的生物环境有关。
Objectives To investigate the mechnisen of increased incidence of liver tumor in the patients with cir- rhosis. Methods Famale BUF rats (6~8 week) inoculated with BUF 7316A hepatoma cells were treated With γTGF-β35ng/day for 14 days. Tumor size was measured from day 5 to 20 after inoculation. TGF- βalso added into mixed lymphocyte cultures (MI.R) and 7316A hepatoma cells cultures. Results Tumor growth in the group of TGF-βtreatement was significantly greater then the control rats (P<0.01). TGF-βwas proved to have inhibitory effect on ivILR (P<0.01) but not on 7316A hepatoma cell prolifer- ation. Conclusion As TGF-βis mostly produced during the liver regeneration and have inhibitory effect on lymphocyte proliferation. It might produce imunological disturbance in the process of liver regeneration and be responsible for increased incidence of liver tumor in the patients with cirrhosis.
