摘要
目的构建转移抑制基因nm23-H_1cDNA克隆,初步分析其mRNA表达与肝内转移、门静脉形成癌栓、TNM分期等临床病理特征的关系。方法利用RT-PCR技术扩增nm23-H_1cDNA片段,并亚克隆人pBluescript Ⅱ SK(-)质粒,借助Northern blot方法,检测10例肝细胞癌及癌旁肝组织nm23-H_1基因mRNA表达水平。结果 nm23-H_1mRNA低表达,诱发肝细胞癌伴肝内转移和/或门静脉癌栓形成的机会增加,nm23-H_1 可能也是肝癌转移抑制基因。结论 nm23-H_1基因mRNA低表达,在肝癌浸润转移过程中起着重要的作用。
Objectives Construct antimetastic gene nm23-H_1 cDNA clone and primarily analyze the relationship between the expression level of nm23-H_1 mRNA and intrahepatic metastasis, portal vein tumor embolus, TNM stage. Methods Amplified nm23-H_1 cDNA fragment by reverse transcription-polymerase chain re- action (RT-PCR) and subcloned into pBluescript Ⅱ SK(-)plasmid. The mRNA expression level was ex- amined in 10 hepatocellular carcinomas and their corresponding liver tissues by Northern blot hybridiza- tion using the above cDNA as a probe. Results The low level of mRNA increased the potential of hepa- tocellular carcinoma (HCC) with intrahepatic metastasis and/or embolus in portal vein. In HCC nm23-H_1 may be an antimetastic gene. Conclusion The low expression of nm23-H_1 gene might play an important role on metastasis and invasion in HCC.
