摘要 目的研究乳鼠心肌细胞的体外培养并进行形态学观察。方法取日龄1~3d Wistar大鼠,胰蛋白酶和Ⅱ型胶原分离消化心肌组织,差速贴壁法纯化心肌细胞,培养于达氏改良伊格尔(DMEM)培养基,显微镜下观察心肌细胞形态及纯度。结果心肌细胞存活率95%,纯度82%,并出现同步搏动。结论胰蛋白酶和胶原酶混合使用可以分离出数量较多、状态良好的心肌细胞。 Objective To study the cultivation and observe the morphological features of myocardial cells of neonatal rats. Methods The ventricular myocardia of neonatal rats with an age of 1-3 days were obtained, and they were digested with pancreatic enzyme and collagenase IL Myocardial cells were isolated with the technique of differential anchoring velocity and they were cultured in DMEM. The morphological feature, survival rate and purity of myocardial cells were observed with light microscopy. Results The survival rate was 95% and purity was 82%. The pulsation of all myocardial cells was synchronous. Conclusion This technique for isolation of myocardial cells is effective, and the situation for cultivation of myocardial ceils is also very satisfactory.
