摘要
通过血凝试验及糖抑制试验,测定褶纹冠蚌的血清及肌肉提取液的凝血活性及糖抑制专一性,同时进行热稳定性、pH及Ca2+影响试验。结果表明,血清和肌肉凝集素都可以凝集多种脊椎动物的红细胞,其中对鸡红细胞的凝集效价均为最高,分别为25和21;血清和肌肉凝集素都不能凝集大肠杆菌,均能凝集嗜水气单胞菌、枯草杆菌、变形杆菌和保加利亚乳杆菌。血清和肌肉凝集素都有一定的热稳定性,血清凝集素60℃处理10min丧失凝集活性,而肌肉凝集素50℃处理10min凝集活性丧失。血清凝集素具有较广泛的pH适应性,在pH6～10内对鸡红细胞均有凝集活力,而肌肉提取液凝集素在pH<7或pH>9时凝集活性消失。血清凝集素能被乳糖和麦芽糖所抑制,肌肉凝集素不能被受试的7种糖所抑制。血清凝集素凝集活性明显地依赖于Ca2+等2价离子,而肌肉凝集素对Ca2+等2价离子没有依赖性。因此,认为褶纹冠蚌血清和肌肉提取液所含凝集素类型不同。用嗜水气单胞菌对褶纹冠蚌进行免疫刺激后,血清和肌肉凝集素对鸡红细胞的凝集效价并没有发生明显变化。
In this study, serum and muscular extraction from Cristaria plicata were detected for agglutination activity and glyco - inhibition. Meanwhile, thermostability and influence of pH and Ca^2＋ on the activity were tested. The results showed that the serum and muscular extraction of Cristaria plicata could agglutinate many kinds of vertebrate erythrocytes, the agglutination titers of the two lectins were the highest for chicken erythrocyte, which were 2^5 and 2^1 respectively, The two lectins could agglutinate Aeromonas hydrophila, Bacillus subtilis, Bacillus proteus and Lactobacillus bulgaricus, and they could not agglutinate E. coll. There were lots of different properties between these two lectins, such as the serum lectins have a higher thermostability that has activity after exposing to 50℃ for 10 minutes, while muscular extraction lectins have no agglutination activity in the same condition. The serum lectins have a pH value range from 6 to 10, while muscular extraction lectins have no agglutination activity when pH 〈 7 or pH 〉 9. The agglutination activity of serum against chicken erythrocyte could be inhibited by lactose and maltose, while agglutination activity of muscular extraction was not inhibited by what had been tested in this study. The agglutination activity of serum depended on Ca^2＋ , while agglutination activity of muscular extraction did not depend on Ca^2 ＋ . The results indicated that the serum lectins and muscular extraction lectins are different in Cristaria plicata. When. Cristaria plicata were infected by Aeromonas hydrophila, no evident changes were found in agglutination titers of the serum and muscular extraction against chicken erythrocytes.
