氧化苦参碱对局灶性脑缺血大鼠脑组织炎症信号TLR4表达的影响 被引量:6

摘要 目的:观察氧化苦参碱对局灶性脑缺血(Focal Cerebral ischemic,FCI)大鼠脑组织Toll样受体4基因、蛋白表达的作用。方法:参照longa等方法制成大鼠局灶性大脑中动脉阻断(MCAO)模型,将80只SD雄性大鼠随机分为假手术组、生理盐水组、氧化苦参碱低、高两个剂量组,分别采用RT-PCR法检测缺血灶周围脑组织TLR4基因、蛋白表达水平。结果:生理盐水组TLR4基因相对表达均明显高于假手术组(P<0.01);氧化苦参碱高剂量组TLR4相对表达量明显低于生理盐水组(P<0.01);TLR4蛋白表达类似于基因相对表达。结论:生理盐水组大鼠缺血灶周围脑组织TLR4基因高表达,氧化苦参碱对TLR4表达有抑制作用,这可能是其抗炎作用的机理之一。 AIM : To observe the effect of oxymatrine on the expression of Toll-like receptor 4 (TLR4) in brain tissue surrounding cerebral ischemic rats. METHODS : The FCI model with thread embolism of middle cerebral artery occlusion (MCAO) in rats was made, 80 male SD rats were divided randomly into shamed-operation, saline and oxymatrine groups, and the oxymatrine group was further divided into two sub-groups which were dealt with the low and high dose of oxymatrine. The expression level of TLR4 and TLR4 mRNA in the brain tissue surrounding cerebral ischemic was determined simultaneously by the usage of immunohistochemical and RT-PCR method respec- tively. RESULTS: The expression of TLR4 gene in saline group was higher evidently than that in shamed-opera- tion group (P 〈 0.01 ), and was obviously lower in oxymatrine group with higher dose than that in saline group( P 〈 0.01 ). TLR4 protein similer to gene. CONCLUSION : Up-regulation of TLR4 expression can be found in sa- line group. Oxymatrine can inhibit the expression of TLR4. The inhibitory effects of oxymatrine on cerebral ischemic may be associated with the inhibition of TLR4 expression.
