摘要
在KNAUER-830型氨基酸全自动分析仪上,OPA柱前衍生法能快速、准确地测定饲料中的氨基酸含量.色谱条件为:流量1ml/min,反应时间5min,柱温45℃,洗脱液为醋酸钠和甲醇混合液,采用梯度洗脱.饲料中氨基酸含量的变异系数在0.37%~7.03%之间.氨基酸回收率除蛋氨酸外,其余都在95%~105%之间.氨基酸含量与峰面积的相关系数在0.9329~0.9997之间.
OPA precolumn derivatization method can rapidly and accurately measure contents of amino acids in feeds on modular KNAUER Amino Acid Analyzer 830. Chromatographic conditions are as follows: flow rate 1 ml/min, reaction time 5 min, temperature of column 45 ℃,gradient buffer is a mixture of anhydrous sodium acetate and methanol. Coefficients of variation of amino acid content is 0.37%~7.03%. Recoveries of amino acid range from 95% to 105% except methionine. Correlation coefficient between amino acid contents and the peak areas ranges from 0.932 9 to 0.999 7.
