摘要
目的观察水蛭素对肝纤维化大鼠肝脏组织结缔组织生长因子(Connective tissue growth factor,CTGF)mRNA表达的影响。方法SD大鼠60只随机分为空白对照组、模型组和水蛭素组共3组,每组20只,用40%四氯化碳(carbon tetra-chloride,CC l4)复制大鼠肝纤维化模型,水蛭素干预12周后处死大鼠,采用实时荧光定量PCR法检测大鼠肝组织CTGFmRNA的表达。结果模型组CTGF mRNA 2-ΔΔC t值(3.59±0.52)显著高于正常对照组(1.02±0.23)(P<0.05),表明模型组CTGF mRNA表达上调。与模型组比较,水蛭素组CTGF mRNA 2-ΔΔC t值(1.83±0.34)显著降低(P<0.05),表明CTGF mRNA表达下调。结论水蛭素可能通过下调CTGF mRNA的表达,抑制肝脏细胞外基质异常增生发挥抗肝纤维化作用。
Objective To observe the effect of hirudin on CTGF mRNA expression in rat liver in hepatic fibrogenesis induced by carbon tetrachloride ( CCl4 ). Methods Studies were conducted in 60 male Sprague - Dawley rats, and they were equally randomly divided into 3 groups with 20 in each, named as blank controls group, model group, and hirudin group. A hepatic fibrosis model on rats was set up with CCl4 (40%). Hirudin were given to the hirudin group fibrotic rats (daily gavage) , normal saline was given to the control group and model group rats. After 12 weeks, CTGF mRNA expression in hepatic tissue were detected by real - time quantitative reverse transcription - PCR. Results The CTGF mRNA expression was up - regulated In model groups, in which CTGF mRNA 2^-ΔΔCt, value were 3.59 ± 0.52 , compared with the blank group ( 1.02±0. 23,P 〈 0.05 ). The CTGF mR- NA expression in hirudin group (1.83± 0.34) was down -regulated compared with the model group( P 〈 0.05). Conclusion - Hirudin may down - regulate expression of CTGF mRNA and Inhibit the abnormal hyperplasia of hepatic extracellular matrix, so it has the function of antihepaticfibrosis.
