摘要
在15L气升式反应器PHB的发酵实验中,进行了欧文氏工程菌Ecc13B-1的高密度培养。实验表明,在无侧孔导流筒的反应器中,发酵53h菌体密度可达57.9g/mL,并使菌体和PHB的生产能力大为提高。发酵中维持稳定的比生长速率有利于提高菌体浓度。结果并说明ALR较传统搅拌罐在高密度培养中更具优势。
The fermentation experiments of recombinant Ecc13B1 for the production of PHB were carried out to achieve high cell density in 15L Air-lift Loop Reactor. The experimental results showed that the cell density can reach 57.9g/cm3 in 53 hours in ALR with holeless draft tube. Furthermore, the production ability of cell and PHB significantly increased. Maintaining the stable specific growth rate in fermentation was helpful to the increase of cell density. The results also proved that ALR was superior to stirred tank for high cell density cultivation.
