摘要
以脱脂米糠为原料,利用响应面法研究了碱性蛋白酶的酶解温度、酶解时间、pH以及酶加量对脱脂米糠蛋白提取率的影响。结果表明回归模型能很好地反映各因素水平与响应值之间的关系,同时得出最佳的提取条件为:温度60℃,时间2.14 h,pH 9.48,酶加量250 U/g。此时理论蛋白提取率为81%,实际测定蛋白提取率为79.84%,蛋白制品纯度为65.32%。
The protein was extracted from defatted rice bran by alcalase hydrolysis. The effects on the protein extraction rate of hydrolyzing temperature, hydrolyzing time, pH and alcalase dosage were studied. The results indicated that the generated regression model represented the relationship between the independent variables and the responses. From the regression model, the optimum conditions of enzymatic pretreatment were identified as follows: hydrolyzing temperature 60 ℃, hydrolyzing time 2.14 h, pH 9.48, and alcalase dosage 250 U/g. Under the optimum conditions, the predicted protein extraction rate was 81% ,whereas the experimental extraction rate was 79.84%. The protein content of finished product was 65.32%.
