摘要
以实验室自行克隆的新基因ZNF191顺序为材料,按三联体密码转换出锌指基因编 码的4个锌指结构域ZNF191a,b,c和d的蛋白质一级顺序,并将它们与已用晶体衍射法确 证了空间构象的7个Cys2His2(C2H2)型锌指进行同源比较,从中发现锌指7ZNF和1ZNF的 一级结构与待检验的锌指顺序同源度高且间隙较小.然后,运用 QUANTA软件包对 4个锌 指结构区 ZNF191a,b,c和d分别进行了计算机三维结构模拟,并以3D profile方法检验了该 结构的准确程度.分析结果提示:ZNF191基因的4个潜在锌指结构区均能够形成正常锌指 结构.
Three dimension conformation of four putative zinc-finger motifs coded by a novel gene ZNF191 cloned in our laboratory was constructed and examined by using the homologybased modeling system. Data of 15 known zinc-finger motifs, 7 of which are C2H2 type,in PDB protein database were selected and used to compare with linear sequence of the zinc-finger motifs to be tested. An alignment was made between the sequence of four putative zinc-finger motifes and that of seven C2H2-type motifs seperately. Using QUANTA software package, each con formation relating to putative zinc-finger motifs was constructed and then examined by 3D profile method. It is shown that all of the putative zinc-finger motifs can form a zincfinger conformation properly.
