摘要
以2'-氯乙基二乙胺盐酸盐(DEAE)为载体修饰剂,对β-硫酸酯乙砜基苯胺(SESA)为载体活化剂,制备多孔球状碱性氨基苯磺酰乙基接枝淀粉(ABSE接枝淀粉)载体,采用重氮化方法固定化葡萄糖异构酶.测定DEAE基团量对固定化葡萄糖异构酶pH-活力的影响,同时对葡萄糖异构酶的最佳固定化条件和性质进行详细的研究.结果表明,DEAE基因虽然并没有使固定化异构酶的最适pH发生移动,但却使其加宽,于pH6仍保持最大活力85%.葡萄糖异构酶经固定化后pH稳定性明显提高,但是温度稳定性却有所下降.该固定化葡萄糖异构酶于4℃储存2个月,活力没有明显改变.
Glucose isomerase was immobilized onto DEAE-(4-amino-benzene sulfonylethyl-porous starch graft copolymers) beads by diazotization. Although the DEAE content did't make the optimum PH of IGI shift, it broadened the pH-activity curve of IGI. The IGI showed improved PH stability as compared to that of free enzyme. When stored at 4℃ for 2 months, no obvious change of IGI activity was found.
