摘要
目的观察PDCFR-a与PDGF-AAmRNA在大鼠不同成熟阶段肺中的表达,以及地塞米松(Dex)对它们在肺内表达的调节。方法溶液杂交法。结果PDGFR-amRNA随胎龄增长而变化;PDCF-AAmRNA随胎龄增加变化不大。Dex能调节大鼠胎肺PDGFR-a的表达,对PDGF-AA无明显影响。结论PDGFR-a、PDGF-AA存在于大鼠肺中,并随胎龄增长而发生变化,提示PDGF与肺发育密切相关;Dex对大鼠肺PDCFR-a的基因表达有调节作用,这可能是激素影响肺成熟的机制之一。
Objective To study o differential expression of p1ate1et-derived growth factor receptor-Q (PDCFR-a) and platelet-derived growth factor-AA (PDCFAA) mRNA levels and regulations of dexamethasone (Dex)on these growth factors, expression in late fatal rat lung development. Methods RNase protection assay wasused to study the expression of message for PDGFAA chain and its cognate receptor-a(PDGFR-Q) during lungdevelopment. whole lung RNA was extracted on l8, l9, and 2l days of gestation (term = 22day), newborn andadult rats. By serum free culture of lung explants, the effects of Dex on PDGFs express ion was also investigated.Results Expression of message for PDGFR-a was low during the pseudoglandular stage on day 18, peakedduring the canalicular stage on day l9 and the saccular stage at day 2l of gestation, fell again in newborn rat,then increased in adult rat; Expression of message for PDCF-AA did not significantly change with advancinggestation; Dex could regulate the expression of mesage for PDGFR-Q in late fetal rat lung. Conclusions Wecondude that the presence of PDGF-AA and PDGFR-Q is compatible with a role for these growth factors during latefetal rat lung. Dex can regulate the gene expression of PDCFR-a in fetal rat lung.
