摘要
目的:研究联用谷氨酰胺和吲哚美辛对重症急性胰腺炎(severe acute pancreatitis,SAP)大鼠胰腺细胞凋亡的影响,并探讨其作用机制。方法:将大鼠随机分成5组:谷氨酰胺治疗组;吲哚美辛治疗组;联合治疗组;重症急性胰腺炎组和正常对照组。术后12、24和36h处死大鼠。取大鼠胰腺病理标本,采用TUNEL法检大鼠测胰腺腺泡细胞凋亡指数。结果:正常对照大鼠胰腺腺泡细胞凋亡指数2.97%。另4组大鼠胰腺腺泡细胞凋亡指数均明显高于对照组,3个治疗组均明显高于SAP组,其中联合治疗组大鼠胰腺腺泡细胞凋亡指数最高。结论:谷氨酰胺和吲哚美辛能促使SAP大鼠胰腺细胞凋亡增加,两药联用有协同作用。
Objective:To investigate the role of glutamine and indomethacin on apoptosis of pancreatic acinar in SAP rats and underling mechanism. Methods:one hundred and four SD rats were randomly allocated to 5 groups:glutamine group(Gin group, n = 24), indomethacin group(IN group,n = 24), glutamine + indomethacin group(G+ I group,n = 24) and SAP group(n = 24). Jejunotomy was performed. Gin group received glutamine, IN group received indomethacin,G + I group received indomethacin and glutamine, SAP group received saline. All groups were subdivided into 3 groups. Eight normal rats were used as controls. At 12,24,36 hours after the induction of SAP. Eight rats in each group were sacrificed 12,24,36hours later. The apoptosis of pancreatic acinar cells was detected by TUNEL method. Results:The apoptotic rate of pancreatic acinar in normal was 2.97 %. The apoptotic rate of pancreatic acinar in the four experimental groups were much higher than that in control group(P〈0.01), among them Gln, IN and G + I were higher than SAP(P〈0.01 ), and highest was occurred in G + I group(P〈0.01 ). Conclusion: Enteral administration of glutamine and glutamine increase the apoptotic rate of pancreatic acinar. There are synergistic effects between Gin and indomethacin.
