注射一株灭活气单胞菌苗对刺参部分免疫因子的影响 被引量:5

Effects of an inactivated bacterium injection on some immune factors in sea cucumber Apostichopus japonicas

摘要 从患有"腐皮综合征"的刺参(Apostichopus japonicas Liao)病灶处分离出1株优势菌株,经形态学、生理生化分析以及16S rDNA分子生物学方法鉴定其为气单胞菌属(Aeromonas)的1个种;人工感染试验证明,该菌株对刺参有一定毒力。将其制备成甲醛灭活菌苗后对健康刺参进行体腔注射,测定灭活气单胞菌苗对刺参部分免疫指标的影响,同时将生理盐水注射组作为对照。结果表明,注射菌苗浓度3.9×108CFU/mL组刺参血清中溶菌酶活力在注射后6h达到峰值,而4.7×107CFU/mL注射组的活力在注射后24h达到最高值;实验组的溶菌酶活力显著高于对照组(P<0.05)。在3.9×108CFU/mL组中,碱性磷酸酶活力于48h达到最高值;而4.7×107CFU/mL组在96h达到最高值,实验组的碱性磷酸酶活力显著高于对照组(P<0.05)。相比之下,注射菌苗对血清的超氧化物歧化酶活力影响却不显著。总体上,该菌苗在一定程度上可以刺激刺参机体产生强的免疫应激反应。注射高浓度菌苗时,溶菌酶、碱性磷酸酶活力达峰所用时间较短而且显著,但其酶活力升高持续时间较短。 The cultured sea cucumber (Apostichopus japonicus Liao) recently suffered from serious disease 'skin ulcer syndrome' which has caused massive losses. One bacterial strain was isolated from these naturally infected sea cucumber, which was the most dominant among the isolates. The bacterium was identified as Aeromonas sp. by morphological, physiological and biochemical tests and 16S rDNA sequence analysis. Challenge test proved that it was the causative pathogen associated with the disease. The bacterium were killed by formaldehyde and used as an inactivated vaccine to inject normal sea cucumber. The immune effects were shown as follows: the highest lysozyme activities were observed at 6 h post-injection in 3.9 ×10^8 CFU/mL group, and at 24 h postinjection in 4.7 × 10^7 CFU/mL group. The alkaline phosphatase (AKP) activities reached maximum at 48 h postinjection in 3.9 × 10^8 CFU/mL group, and at 96 h post-injection in 4.7 ×10^7 CFU/mL group. However, there was no significant difference in superoxide dismutase (SOD) between the experimental groups and the control group. In general, the inactivated bacterium could increase the immune effects on sea cucumber. In addition, as the concentration of the inactivated bacterium was higher, lysozyme and AKP activities could reach the maximum rapidly and significantly. But the increased activities could not last long.
