摘要
目的研究基质金属蛋白酶-9(matrix metalloproteinase-9,MMP-9)在大鼠茄病镰刀菌性角膜炎中的表达,分析MMP-9与真菌性角膜炎发病的关系,为针对抑制MMPs活性治疗真菌性角膜炎提供理论依据。方法角膜划痕法联合角膜接触镜法制作大鼠茄病镰刀菌性角膜炎动物模型;应用实时荧光定量聚合酶链式反应(Real-time polymerase chain reaction,Real-time PCR)和免疫组织化学染色检测MMP-9在mRNA水平和蛋白质水平的表达。结果正常大鼠角膜组织中有MMP-9 mRNA的表达,MMP-9分布于角膜上皮层的少数基底细胞;茄病镰刀菌感染角膜后,MMP-9 mRNA表达明显上升,MMP-9主要分布于浸润的炎症细胞。结论MMP-9的表达与真菌性角膜炎角膜病变的严重程度呈正相关,MMP-9在真菌性角膜炎的发病中可能具有重要作用。
Objective To study the relationship between fungal keratitis and matrix metalloproteinase-9 (MMP-9) by investigating the expression pattern of MMP-9 in Fusarium solani (F. solani) keratitis rats, so as to offer the possibility of treatment of fungal keratitis by inhibiting MMPs on theory. Methods Experimental model of F. solani keratitis was built by the combination methods of the corneal scarification and hand-made contact lens in rat. Expression of MMP-9 both in mRNA level and protein level were explored by the methods of real-time polymerase chain reaction (PCR) and immunohistochemistry. Results MMP-9 both in mRNA level and protein level were expressed in the normal cornea and localized in a few corneal basal cells. MMP-9 expression of mRNA and protein were significantly raised up after the cornea infected by F, solani and mainly localized in infiltrated inflammatory cells. Conclusion Expression of MMP-9 is positively correlated with the extent of F. solani infection. MMP-9 may play an important role in the process of fungal keratitis.
