摘要
目的:观察凋亡蛋白p53、Bax、Bcl-2,粘附蛋白整合素α3β1、肌营养不良蛋白聚糖(DG)在Heymann肾炎鼠肾小球的表达,以及1,25-(OH)_2D_3对其表达的影响。方法:雌性SD大鼠随机分为对照组和Heymann肾炎模型组;后者经腹腔注射FX1A抗原诱导肾炎模型,再随机分Heymann肾炎组和1,25-(OH)_2D_3组。1,25-(OH)_2D_3组皮下埋置渗透性微量泵,给予1, 25-(OH)_2D_33ng·100g^(-1)·d^(-1),连用4周。检测大鼠尿蛋白、尿足细胞。电镜检测每视野足细胞数、足突平均宽度。免疫组织化学SP法观察肾小球p53、Bax、Bcl-2,整合素α3β1、肌营养不良蛋白聚糖(DG)蛋白的表达。原位末端标记(TUNEL)法检测足细胞凋亡。结果:与对照组相比,Heymann肾炎组大鼠尿蛋白、尿足细胞排泄明显升高;足细胞数目减少,足突宽度增加;肾小球p53、Bax表达增高,Bcl-2表达减少,足细胞凋亡明显增多;整合素α3β1、DG表达明显减少。与Heymann肾炎组相比,1, 25-(OH)_2D_3组尿蛋白、尿足细胞排泄明显减少;肾小球p53、Bax表达减少,Bcl-2表达增加,足细胞凋亡减少;整合素α3β1、DG表达明显增加。结论:1,25-(OH)_2D_3可减少Heymann肾炎鼠肾小球p53、Bax的表达,增加Bcl-2的表达,减少足细胞凋亡。亦可增加整合素α3β1、DG在肾小球中的表达,减少足细胞脱落。从而维持肾小球足细胞数量。
To observe the effect of 1,25-(OH) z D3 on apoptotic protein p53, Bax, Bcl-2 and attachment protein integrin α3β1,dystroglycan(DG) expression in glomeruli in rats with passive Heymann nephritis(PHN). Method: SD rats were randomly divided into two groups:control group,PHN model group which was induced by antigen(FX1A) peritoneal injection. After these models were successfully established,the), were randomly divided into two groups:model group without therapy (Heymann group) and 1,25- ( OH ) 2 D3 group with 1,25- ( OH )2D3 therapy. 1,25- ( OH ) 2 D3 group was embedded with the osmotic minipump for giving 3 ng·100g^-1·d^-1,25- ( OH ) 2 D3. After the treatmemt with 1,25- ( OH ) 2 D3 for 4 weeks, proteinufia, urinary podoctye ( UPC ) were measured. Average numbers of podocytes per field and mean of the foot process width (FPW) were detected under electron microscopy. The expression of p53, Bax, Bcl-2,integrin α3β1, DG in glomeruli was observed by immunohistochemical SP methods. The apoptosis podocyte was detected by the transferase-mediated dUTP nick-end labeling (TUNEL) assay. Result: Compared with control group, the level of urine protein, UPC in Heymann group was significantly increased; average numbers of podocytes per field was dramatically reduced; FPW was increased; the expression of p53,Bax protein increased, Bcl-2 decreased in glomeruli,which increased podocyte apoptosis. Compared with Heymann group,the level of urine protein, UPC in 1,25-(OH)2D3 group was significantly decreased; the numbers of podocytes per field of vision was increased; FPW was reduced; the expression of p53, Bax protein decreased, Bcl-2 increased, which decreased podoctye apoptosis. Conclusion: 1,25-(OH)2D3 could contribute to decrease p53 ,Bax and increase Bcl-2 expression in Hey- mann rats,which reduce podoeyte apoptosis. Furthermore, 1,25-(OH) 2 D3 increased integrin α3β1, DG expression, which prevented podoeyte attachment. So preserve podoeyte numbe including inhibiting apoptosis and detachmen.
