核转录因子抑制蛋白α突变体对大鼠急性胰腺炎的保护作用

Protective effect of IκBαM mutant on the acute pancreatitis in rats

摘要 目的观察核转录因子抑制蛋白α突变体(hcBαM)对大鼠急性胰腺炎的保护作用。方法蛙皮素诱导建立SD大鼠急性胰腺炎模型。分4组:空白对照组A组、急性胰腺炎组B组、转染PcDNA3.0组c组、转染PcDNA3.0-IκBαM组D组。分别于24h检测各组血清肿瘤坏死因子α(TNF-α)、淀粉酶的表达;逆转录-聚合酶链反应(RT—PCR)检测胰腺组织中Fas、FasL、Caspase-3mRNA的表达;组织学检测胰腺腺泡细胞坏死情况。结果D组与B、C组比较:血清TNF-α、淀粉酶表达水平明显降低,差异有统计学意义[TNF-α(587±93比915±102;587±93比897±116);淀粉酶(6773±663比8973±825;6773±663比9021±763),P〈0.05];胰腺组织Fas、FasL、Caspase-3mRNA表达明显升高,差异有统计学意义[Fas(0.572±0.103比0.473±0.084;0.572±0.103比0.468±0.071);FasL(0.627±0.124比0.397±0.084;0.627±0.124±0.391±0.072);Caspase-3(0.893±0.142±0.513±0.122;0.893±0.142比0.508±0.130),P〈0.05];胰腺组织腺泡细胞坏死明显减少,差异有统计学意义(P〈0.05)。B、C、D组与A组表达差异有统计学意义(P〈0.05)。结论IκBαM通过诱导胰腺腺泡细胞凋亡对大鼠急性胰腺炎发挥保护作用。 Objective To study the protective effect of the IκBαM mutant gene on the acute pancreatitis (AP) in rats. Methods The pancreatitis in rats were induced by cerulean (CR), and the rats were randomly divided into group A (control) ,group B (AP) ,group C (AP + PcDNA3.0 transfection), and group D ( AP + PcDNA3.0-IκBαM transfeetion). The expression of TNF-α, Fas, FasL, and Caspase-3 was detected by ELISA and RT-PCR. The necrosis of acinar cells was evaluated by immunohistochemistry. The serum level of amylase in every group was determined. Results In comparison with groups B and C, the expression of TNF-α and the serum level of amylase were decreased significantly, the expression of Fas, FasL, and Caspase-3 mRNA was increased significiantly, and the number of necrotic acinar cells was decreased significantly in group D. All pancreatitis groups had significant difference with group A ( P 〈 0. 05 ). Conclusion The IκBαM can protect against the acute pancreatitis in rats by inducing the apoptosis of the acinar cells.
