摘要
本文主要探讨了大腹园蛛与虎纹捕鸟蛛丝在不同溶剂里的溶解情况,及其再生丝蛋白溶液静电纺纤维的形态结构与分子构象。结果表明,大腹园蛛丝的溶解率明显高于虎纹捕鸟蛛丝,这是它们的结晶度和氨基酸组成的差异造成的。以六氟异丙醇(HFIP)为溶剂时,再生大腹园蛛丝(AVSH)、再生大腹园蛛丝蛋白凝胶(AVGH)以及虎纹捕鸟蛛丝(OHSH)溶液的静电纺纤维直径的大小依次是AVSH<OHSH<AVGH,而以甲酸和HFIP为混合溶剂时,再生大腹园蛛丝蛋白溶液(AVSHF)的静电纺纤维直径又小于AVSH,且比后者要均匀得多。红外光谱的测定结果表明,AVGH静电纺纤维中β-折叠分子构象的含量少于AVSH纤维,溶剂对再生蛛丝蛋白静电纺纤维的分子构象无显著影响。
The solubility of Ornithoctonus huwenna and Araneus ventricosus spider silk in different solvents was measured, and the microstructures, molecular conformations of their electrospun regenerated fibers were studied in this article. The results show that in HFIP the silk of Araneus ventricosus spider has much higher solubility than that of Ornithoctonus huwenna, which is attributed to the crystal degree and amino acid composition. Diameters of electrospun fibers from regenerated Araneus ventricosus spider silk(AVSH), Araneus ventricosus spider silk gelatin(AVGH) and Ornithoctonus huwenna spider silk(OHSH) solutions in hexaflorisopropanol (HFIP) are in the following sequence of AVSH 〈 OHSH 〈AVGH. On the other hand. when the solvent is the mixture of formic acid and HFIP, the diameter of electrospun regenerated Araneus ventricosus spider silk(AVSHF) fiber is finer and more even than that of AVSH. The results of FT-IR indicate that there is less β-sheet structure in AVGH eleetrospun silk than AVSH, and solvents have no notable effect on the molecular conformation of electrospun regenerated spider silk.
