摘要
目的建立较为全面反映肝星状细胞活化效应的细胞模型。方法应用脂多糖刺激体外培养的大鼠HSC。MTT法测定HSC增殖率。免疫组化法显示HSC表达Ⅰ、Ⅲ型胶原水平,并进行图像分析。结果LPS明显刺激HSC的增殖和提高其Ⅰ、Ⅲ型胶原表达水平,与对照组比较有非常显著性差异(P<0.01)。结论LPS诱导HSC活化模型能够相对全面反映HSC活化效应。
Objective To develope cellular model of HSC activation. Methods Isolated rat HSC cultured in vitro was stimulated with lipopolysccharide（LPS）. Proliferation of rat HSC was detected with MTT technique. Expression levels of collagen Ⅰ and collagenⅢ in rat HSC were showed with immunochemistry technique and analyzed with image analysis system. Results LPS significantly promoted proliferation elevated expression levels of collagen Ⅰ and collagen Ⅲ in rat HSC. Conclusion Model of rat HSC activation stimulated with LPS can reflect more intact HSC activation effects.
