摘要
目的:探讨过表达maspin基因对肝癌细胞株MHCC-97中VEGF和uPA表达的影响及意义.方法:构建maspin/pEFIRES-N表达质粒,重组质粒转染后,采用RT-PCR及Western Blot方法分别从mRNA和蛋白水平检测肝癌细胞株MHCC-97中maspin,VEGF和uPA转染前后的表达.结果:成功构建maspin/pEFIRES-N重组质粒,并稳定转染到肝癌细胞株MHCC-97中.比较maspin/pEFIRES-N组和pEFIRES-N组的肝癌细胞株MHCC-97中maspin,VEGF和uPA的表达,结果显示转染maspin基因后在肝癌细胞株MHCC-97中maspin的mRNA和蛋白表达明显增强,而VEGF,uPA的mRNA和蛋白表达明显减弱(P<0.05).结论:在肝癌细胞株MHCC-97中,maspin基因的过表达会降低VEGF和uPA的mRNA及蛋白表达,从而可能抑制新生血管形成、阻断纤溶酶原激活物调节uPA的活性,减少ECM的降解,阻止肿瘤细胞的转移.
AIM:To explore the influence and significance of maspin overexpression on the expressions of VEGF and uPA in hepatocareinoma cell MHCC-97. METHODS: We constructed maspin/pEFIRES-N expression plasmid, and detected the changes of maspin, VEGF and uPA at the levels of mRNA and protein in hepatocarcinoma cell MHCC-97 before and after the transfection by RT-PCR and Western Blot, respectively. RESULTS: The maspin/pEFIRES-N plasmid was constructed successfully, and was stably transferred into MHCC-97 cells. The expressions of maspin mRNA and protein increased in hepatocarcinoma cell MHCC-97 after the transfection of maspin/pEFIRES-N plasmid, while the expressions of VEGF, uPA mRNA and protein decreased compared with those after the transfection of pEFIRES- N plasmid ( P 〈 0. 05 ). CONCLUSION: In hepatocarcinoma cell MHCC-97, maspin overexpression decreases the expressions of VEGF and uPA, which inhibits neovascularization, blocks the activity of uPA regulated by activator of plasminogen.
