口腔鳞癌中VCAM-1基因表达与临床病理学特征及血管生成的关系 被引量:3

Correlations between the expression of vascular cell adhesion molecule-1 gene and clinical pathological characteristics and angiogenesis in oral squamous cell carcinoma

摘要 目的:研究口腔鳞癌中血管细胞黏附分子-1(vascular cell adhesion molecule-1,VCAM-1)的基因表达及其与临床病理学特征和微血管密度(microvessel density,MVD)的关系。方法:应用组织原位杂交和免疫组化检测48例口腔鳞癌组织和10例正常口腔黏膜组织中VCAM-1mRNA和VCAM-1蛋白的表达和定位,并计数微血管密度(MVD)。采用SPSS13.0软件包中的t检验及χ2检验进行统计学分析。结果:VCAM-1mRNA主要定位于肿瘤细胞胞质,VCAM-1蛋白主要定位于肿瘤细胞胞膜和胞质;VCAM-1mRNA和蛋白在口腔鳞癌中的表达均显著高于正常口腔黏膜组织(P<0.01);VCAM-1的表达与口腔鳞癌的浸润深度、淋巴结转移密切相关(P<0.01),而与患者的年龄、性别及病理分级无关;VCAM-1的表达与MVD值呈正相关(P<0.01)。结论:VCAM-1的高表达可能在口腔鳞癌的侵袭和淋巴结转移过程中起着重要作用,并与肿瘤血管生成有关。 PURPOSE: To investigate the correlations between the expression of vascular cell adhesion molecule-1 (VCAM-1) gene and clinicopathologic characteristics and microvessel density (MVD) in oral squamous cell carcinoma (OSCC). METHODS: Expression and location of VCAM-lmRNA and protein in 48 OSCCs and 10 normal controls were detected by in situ hybridization and immunohistochemical staining,MVD was also assessed. Statistical analysis was performed using SPSS13.0 software package for Student's t test and Chi-square test. RESULTS: VCAM-lmRNA was mainly detected in cytoplasm of OSCC.VCAM-1 protein was mainly detected in membrane and cytoplasm of OSCC. The expression of VCAM-lmRNA and protein were significantly higher in OSCC tissues than normal oral tissues (P〈0.01). In the clinicopathologic factors, lymph node metastasis and depth of infiltration were closely correlated with VCAM-1 expression (P〈0.01), but there was no significant correlation between the positively expression and patients'sex, age and tumor' differentiated degree (P〉0.05). CONCLUSIONS: Overexpression of VCAM-1 gene in OSCC may play a potential role in the development of OSCC.h is closely associated with lymph node metastasis and the angiogenesis.
